All the hundreds of inactivated Viral Transport Medium(Inactivated) products on the market can get correct results?

No! The detection results are greatly related to the guanidine concentration in the lysate.High concentration of guanidinecan effectively inactivate the virus and preserve nucleic acid.But, excessive guanidine residue during nucleic acid extraction inhibited enzyme activity in PCR system.
Low concentration of guanidine, unable to inactivate the virus and preserve nucleic acid effectively risk to the safety of the testers.

The design concept of YOCON Viral Transport Medium(Inactivated)

In the process of product design, we added appropriate concentration of guanidine and other components to promote virus lysis, which could not only ensure the inactivation effect of virus, but also avoid the influence on the later fluorescence quantitative PCR process. There was no significant difference between the results obtained from the inactivated virus sample tube and those obtained from traditional non-inactivated virus sample product.

Can nucleic acids be detected using Viral Transport Medium(Inactivated)?

100ul of influenza virus was added to 1ml of non-inactivated and inactivated sampling solutions respectively. After being mixed and placed at 4℃ and 25℃ for 24 hours respectively, nucleic acids were extracted for RT-qPCR. The amplification curve and Ct value of each group were as follows:

The virus was mixed with the Viral Transport Medium(Inactivated) and then been placed at 25℃ for 7×24h,after which the nucleic acid was extracted and tested. The Ct value was slightly increased, indicating that the detection results were partially affected.

After being placed at 4℃ and 25℃ for 24 hours, there was no difference in nucleic acid test results between the two products. After being placed at 25℃ for 7×24h, the expression of virus can still be detected in the inactivated sample tube product.  

What's the result of virus inactivation using an inactivated sampling tube? 

Viral Transport Medium VS Viral Transport Medium(Inactivated) 

1mL of influenza virus was added into 1ml of Viral Transport Medium and Viral Transport Medium(Inactivated) respectively, mixed and placed at 25℃ for 15 minutes, and then PEG solution was used for precipitating the viruses, followed by PBS washing, after which the sediment was resuspended. The resuspended suspension was infected with MDCK cells, and 3 days later, we proceed as follows: 1. We observe the cytopathic conditions under the microscope; 2. The supernatant was collected to extract nucleic acid, and RT-qPCR was used to detect the nucleic acid content of the virus in the cultured supernatant.

2.Virus nucleic acid content detection: The amount of viral nucleic acid in the Viral Transport Medium(Inactivated) group was much lower than that in the Viral Transport Medium treatment group.

The inactivation effect of virus treated with inactivated sampling solution is remarkable.
